Metrology for innovative nanotherapeutics

WP2: Traceable methodologies for nanoparticle characterisation

Main goals:

Develop and harmonise traceable methodologies for measuring:

  1. particle size and concentration of small (diameter < 10 nm) MONPs and their typically non-spherical aggregates and/or agglomerates;
  2. lipid composition, RNA loading and structural integrity of lipid-based nanoparticles;
  3. surface properties, including the amount, chemical composition, and homogeneity of the surface coating.

 

 

Techniques:

  1. Traceable size and concentration measurements of small MONPs by AFM-SEM-EDX and SAXS;
  2. Lipid composition of LNPs and liposomes by LC-MS/MS and Raman micro-spectroscopy
  3. RNA loading of lipid-based NPs by HPLC-UV and capillary electrophoresis (CE-LIF)
  4. Structural properties and structural integrity of the LNPs and liposomes by complementary approaches including SAXS, WAXS, SANS and cryo-EM coupled with/without RNA staining;
  5. Characterisation of surface coatings by DSC, TGA, UHPLC-HRMS/MS, qNMR, SAXS, SANS and 3D-cryo-TOF-SIMS;
  6. Data handling and harmonisation

Short bio: Dr. Robin Schürmann is a senior scientist in X-ray radiometry at the Physikalisch-Technische Bundesanstalt (PTB). He holds a PhD in chemical physics on the physicochemical mechanisms of radio-enhancement in cancer therapy from the University of Potsdam and has broad expertise in the characterisation of nanoparticles and surfaces.He is an expert in traceable measurements of nanomaterials with synchrotron radiation
